The most popular screen resolution is still 1024 x 768, although larger monitors are gaining a foothold. For example I've got 3 monitors in my office all of them with resolutions higher than than 1024 x 768.

In general you'll want to cover as many screen resolutions as possible, including smaller ones such as 800 x 600. Designing using fluid width as opposed to fixed width will help in this, although you'll possibly have to make a compromise on lower resolutions.

There are tools to help you simulate different screen resolutions. The IE developer toolbar can force your browser to 1024 x 768 - it's not perfect, but it's close to. There are similar tools for FireFox.
